Method to determine unfertilised eggs of drosophila Method to determine unfertilised eggs of drosophila / 2487934
Undeveloped eggs are placed for 45-50 minutes in a four percent solution of sodium hypochlorite (NaOCl), and on the basis of the number of dissolved eggs they define the quantity of unfertilised eggs. The proposed method makes it possible to perform mass investigations of quite large samples, to detect specifics at the level of frequency of occurrence of unfertilised eggs, both of genetically dependent factors and stresses of the environment of various nature at the reproductive system of the drosophila. Besides, the proposed method of chorion removal makes it possible to detect a share of parthenogenetically developing eggs, which is specific for certain types of drosophila, and also disturbances in the reproductive system of drosophila males. The method makes it possible to process quite a high quantity (100-200 pcs.) of undeveloped eggs referred to early embryonal parts within a short period of time, makes it possible to identify sterility of males. With the help of this method both for parent and several next generations of drosophila, they determine a prolonging effect of biological exposure of a stress agent at ability to leave posterity.

Additionally, it is important to carry out iridoscopy for the lambs aged 5-mo along with detection of color purity - the absence of stripes, lines, grooves, lacunes, spots and homogeneity - the absence of contrast distinctions in pigmentation of iris color. Then comes the selection of stud rams at steady distribution of pigment without any variations of color tone along the whole surface of iris and along its all projectional areas of the right and left eyes in 12.00-11.59 sector clockwise. This invention relates to the field of agriculture, particularly poultry, and can be applied for the prevention of technological stress in laying hens egg breeds in the critical periods of development of the chicken embryo.

It is known that the effect of technological stress leads to the activation of free-radical processes [4] and increases lipid peroxidation in the tissues of chicken embryos, which causes significant economic damage to the poultry industry [1, 2]. It is therefore important prevent these phenomena using environmentally safe natural metabolites [3, 5]. The sodium salt of a-lipoic acid for the treatment of hatching eggs is unknown. Lipoic acid and its salts actively used in medicine, but in agriculture, its use is extremely small. Sodium salt of lipoic acid is involved in the oxidative decarboxylation of pyruvic acid and a - ketoacids, in the process of formation of energy in the body, in the regulation of lipid and carbohydrate metabolism, has a lipotropic effect, affects the metabolism of cholesterol, improves liver function [6]. The sodium salt of lipoic acid due to its good solubility in water, the lipoic acid is insoluble in water. It should be noted that this metabolite has never been used to prevent those of the ideological stress in poultry, as well as its influence on the chicken embryo is not known.

The purpose of the invention is the prevention of technological stress in laying hens egg breeds in the critical periods of development of the chicken embryo by reducing processes lipoperoxidative, optimization of metabolism.

The technical result is stimulation of embryonic development, egg chickens, increase output conforming chickens, prevention of technological stress.

This result is achieved by reducing processes lipoperoxidation and optimize the level of metabolism during critical periods of development of the chicken embryo, by means of a single processing of hatching eggs, namely, before placing in the incubator aerosol spray on the surface of egg shells cause a 0.01% solution of sodium salt of lipoic acid.

The method is as follows:

On the surface of the shell for 3-4 hours prior to incubation of the spray is applied an aqueous solution of sodium salt of lipoic acid in a concentration of 0.01%. Previously the drug is dissolved and mixed in distilled water at a temperature of 18-22°C.

Example 1. Eggs of the cross "shaver Braun was received from one parent stock, sorted by 306 pieces in each batch according to the principle of analogues with respect to time of the demolition and retention and weight. On the surface of the shell struck in the form of AE is the open air the solution under the above scheme. As control group were selected eggs of the same batch on the principle of analogues, which were processed according to the technology adopted in the economy. The obtained data are presented in table 1. The table shows that the conclusion of an experimental batch exceeds the control 11,77%and hatchability on 11,03%. Significantly decreased such waste incubation: "frozen" 4 times, "Sadochok" 3 times, the "weak" 5.1 times.

Example 2. The laying of the eggs of the cross "shaver Braun" on the incubation of the test and control groups was 05.03.2012, at 12 o'clock. 25.03.2012, began to withdraw, the chord continues 29 hours, which is 9 hours ahead of the normal output. Thus, the development of the allantois in the experimental group on the 11th day ahead of control that can be seen from table 2.

As can be seen from table 5 the drug has antioxidant action in the body of the chicken. The table shows that the intensity of peroxidation in experimental group is lower than in the control. Thus, the secondary product of lipid peroxidation in the form of malonic dialdehyde (MDA) decreased by 12.5%. Also in reducing the intensity of the processes of lipoperoxidation, increased antioxidant enzymatic defenses of day-old Chicks, which resulted in the increase of superoxide dismutase (SOD) in 1.5 times. All bio is himicheskie indicators were within the physiological norm.

Thus, on the basis of the performed experiments, there is an increase in the intensity of output with high viability young for a long period of postembryonic development. When this chick in an experimental batch was higher on 11,77% (p<0,001), and hatchability of eggs on 11,03% (p<0,001) compared with the control group.
