Method to determine unfertilised eggs of drosophila Method to determine unfertilised eggs of drosophila / 2487934
Undeveloped eggs are placed for 45-50 minutes in a four percent solution of sodium hypochlorite (NaOCl), and on the basis of the number of dissolved eggs they define the quantity of unfertilised eggs. The proposed method makes it possible to perform mass investigations of quite large samples, to detect specifics at the level of frequency of occurrence of unfertilised eggs, both of genetically dependent factors and stresses of the environment of various nature at the reproductive system of the drosophila. Besides, the proposed method of chorion removal makes it possible to detect a share of parthenogenetically developing eggs, which is specific for certain types of drosophila, and also disturbances in the reproductive system of drosophila males. The method makes it possible to process quite a high quantity (100-200 pcs.) of undeveloped eggs referred to early embryonal parts within a short period of time, makes it possible to identify sterility of males. With the help of this method both for parent and several next generations of drosophila, they determine a prolonging effect of biological exposure of a stress agent at ability to leave posterity.

Additionally, it is important to carry out iridoscopy for the lambs aged 5-mo along with detection of color purity - the absence of stripes, lines, grooves, lacunes, spots and homogeneity - the absence of contrast distinctions in pigmentation of iris color. Then comes the selection of stud rams at steady distribution of pigment without any variations of color tone along the whole surface of iris and along its all projectional areas of the right and left eyes in 12.00-11.59 sector clockwise. The invention relates to the field of agriculture and in particular to the breeding and genetics of cattle.

The known method of selection of cattle for milk production (EN 2391815, the date of publication of the application 20.06.2010).

The known method of selection of cattle for meat production (EN 2366170, publication date 10.09.2009), consisting in the determination of erythrocyte antigen markers of higher and lower energy growth in heifers Simmental breed. If the genotype of the animal markers of increased energy growth or dominance over the antigens of low-energy growth make the selection.

The disadvantage of these methods is the detection of antigen markers productivity in rocks of the milk and dairy beef cattle that are not markers of beef cattle productivity, in particular, the Kalmyk breed.

The proposed method is based on the use of red blood cell antigens. Antigens are passed from parents to their descendants as hereditary units. The synthesis of each antigen due to the action of a single gene.

In this regard, erythrocyte antigens and specific complexes represent the elements of the genotype, which is due to the characteristics of the organism, including those related to its productive qualities. Analysis of erythrocyte antigens POS who s to judge the degree of participation of the inheritance of the father, mother and more distant ancestors in the formation of the genotype of the offspring. On this basis, we investigated their breeding value as a marker genotype in the study of inheritance of economically-useful qualities.

Disclosure of inventions

How is that 6-months of age revealed in the blood of the presence of erythrocytic antigen markers productivity tall type: G2 E'3 R2. If the genotype of the animal markers productivity tall type produce a selection of animals.

Selection - save the animals adapted to the environment, or meet the requirements of a person while removing animals that are not adapted to the conditions of the external environment.

Each breed of cattle are heterogeneous in productivity, morphological and physiological characteristics of animals that make up a kind of group - intra-breed types.

Animals of different intra-breed types with their distinctive biological and economic characteristics expands the possibilities for further improvement of breeds, enables you to identify potential opportunities to increase productivity.

The increase of livestock production must be due to the intensification, which is based on the selection and breeding work. Perfection is the study of existing and creation of new breeds, types and lines of beef cattle is one of the most important tasks of the selection on the modern stage. In order to accelerate the selection along with the traditional methods, you must use the new genetic methods can greatly increase the level of effectiveness of the breeding work.

The main object of experimental studies was the herd Kalmyk breed pedigree reproducer JSC "50 years of October" October district of the Republic of Kalmykia, number 230 goals. Research material as well as documents zootechnical and pedigree records were used immunological card of genetic certification of breeding animals, made on the basis of an analysis of a blood in the laboratory of immunogenetics of JSC "Kalmyk" breeding RK.

Studies were conducted on the bulls Kalmyk breed. To identify features of the genotype steers of different measurements and live weight at 18 months of age, animals were divided into two groups. In the first group consisted of bulls, with the height at the withers more than 110 cm (tall type), gobies, having smaller height at the withers, were assigned to the second group (compact type). All were 20 and 16 calves in each group, respectively. Other measurements in different age periods were studied on 5 extraction of the am of each type.

In zootechnics the question about the reliability (materiality) differences plays an important role. The reliability of difference means the difference between the selected indicators entails differences between the respective General indicators. If it turned out that the difference unreliable, then this means that conclusions about the difference between the sample results cannot be transferred to the General indicators, as they can vary or be the same.

The results showed that the difference between groups at 8 months was reliable with P>0,99. Live weight of bulls compact type was greater than 3.2 kg in 8 months (when registered with the experience).

To 18-month age bulls compact type yielded tall type height at the withers by 2.64 cm and the height at the sacrum by 2.44 cm, the difference in live weight was 22.6 kg P>0,95. Therefore, gobies tall type to 18 months "surpassed" in live weight of steers compact type (table 1).

Table 1

At screening, all calves were examined for antigenic composition of blood with establishing authenticity of origin (table 2).

The frequency of occurrence of erythrocyte antigens varies Dov is correctly significantly.

In the a-system antigens A1/A2 most common in type 1 cattle (frequency - 0,40%) compared with the second type (frequency -0,063%).

In the system, the high - antigens G2 (0,50%),'(0,60%), D'(0,65%), E'3(0,85%) in type 1 than in 2 - 0,063%, 0,25%, 0,25%, 0,13% respectively. Other alleles At the locus (O4, E'1, I') are the same as for the first type and the second.

In the S-locus in type 1 high frequency antigens C1 (0,35%) and C2 (0,55%) compared with the second - 0,13% and 0.31%, respectively. It should be noted that the antigen R2 was observed only with the first type of frequency of occurrence of 0.80%. Antigen x2 occupies an intermediate position.

Systems-F - I-Z - frequency antigens F/V Z was the first type is 0.55%, the second - 0,25%.

As can be seen from the table, there are antigens that are responsible for visokorolski. As well as tall gobies significantly greater live weight at 18 months of age compact Bychkov, we can conclude that these antigens G2 E'3 R2 is also responsible for live weight.

The proposed antigens G2 E'3 R2 be considered markers of productivity in 18 months of age, selection for increased body weight by taking into account antigenic factors.
