The method for preparing the saponin-containing extract containing pre-soaking of Saponaria officialis L. roots in distilled water, ultrasonic extraction, filtration in the specific environment.

EFFECT: method provides higher quality of the end product and reducing a time for preparing the saponin-containing extract from Saponaria officinalis L.

The invention relates to the chemical-pharmaceutical and medical industry, namely to a method of obtaining biologically active substances from plant raw materials.

A method of obtaining saponins ivy (RF Patent №2395516, C07H 1/08, C07H 15/256, A61K 36/25, priority 18.12.2008) by extraction of the containing vegetable raw materials in airtight conditions in the aquatic environment or in an environment of water 0,5-5% ammonia solution at a temperature of 110-200 C.

The main disadvantages of this method are conducting extraction at high pressure and high temperature, which significantly increases the cost of the extraction process, and the use of ammonia as a solvent.

Also a method of obtaining saponins soybean (Patent KR №20030059028, C12P 19/44, priority 13.06.2003), which implies that water extraction and use of treated by aqueous extract of the enzyme beta-glucosidase.

The disadvantages of this method is the low yield of the desired product and the use of expensive enzyme for the treatment of the extract.

Closest to the proposed method is a method of ultrasonic extraction of saponins from the roots of ginseng (J. Wu, L. Lin, F. Chau Ultrasound-assisted extraction of ginseng in saponins from ginseng roots and cultured ginseng cells. Ultrason. Sonochem. 2001. Vol.8. P.347-352), based on the destruction of the internal cellular structure and release the contents of the cells to accelerate mass transfer. The effect is achieved by fast selective heating of raw material in the solvent, partially transparent to the mechanical action of acoustic cavitation.

The disadvantage of this method is the use of organic solvents and duration of extraction (>2 hours).

The objective of the invention is intensification of the process of obtaining capodistrias extracts and reduction of the costs of extraction.

The technical result of the invention is to increase the quality of the desired product using non-toxic solvent (water)and reducing the time of receipt capodistrias extracts from Saponaria officinalis L.

The task and this technical result is achieved by the fact that a method for obtaining capodistrias extracts, including soaking roots Saponaria officinalis L. in distilled water, ultrasonic extraction, filtration extract from vegetable raw materials, according to the invention, the extraction was performed under the influence of ultrasound with the frequency of 30 kHz and the intensity of 228% to 232 W/cm 2 , for 19% to 21 minutes with preliminary soaking raw materials in distilled water for 19% to 21 minutes. The ultrasound is performed at a temperature of 25 C with a ratio of vegetable raw materials and water 1:5.

The use of ultrasound with the frequency of 30 kHz allows to considerably reduce the time needed to obtain capodistrias extracts. When you use ultrasound with frequency below 30 kHz indicators capodistrias extracts worse, the use of ultrasound with the frequency of above 30 kHz impractical due to excessive energy consumption.

The use of ultrasound intensity 228% to 232 W/cm 2 provides the reduction of time of extraction. When using intensity of ultrasonic affecting is less than 228 W/cm 2 increases the time to achieve optimal yield and increase in intensity over 232 W/cm 2 leads to the deterioration get capodistrias extracts.

Time of ultrasonic influence 19 to 21 minutes is optimal for obtaining capodistrias extracts with the best results (in dry substance and foam-making abilities). Reducing the time of exposure of less than 19 minutes is not possible to achieve optimum output of the extract and the increase of time of ultrasonic processing more than 21 minutes leads to the destruction allocated saponins.

Process temperature (25 C) selected experimentally, at a given temperature optimizes performance capodistrias extracts.

Pre-soaking of plant raw materials (19% to 21 minutes) selected experimentally, as this time soaking optimizes performance capodistrias extracts.

The proposed ratio of vegetable raw materials and water is the most appropriate, as this ratio provides the most complete removal of saponins from vegetable raw materials.

The method of obtaining capodistrias extracts was performed as follows. Hanging plant materials were placed in chemical glass and filled with distilled water (water duty 1:5), after which the raw materials were left to soak for 19% to 21 minutes. Ultrasonic generator tuned according to the intensity, then cap the ultrasonic generator immersed in a glass of vegetable raw materials and conducted the processing of raw materials. After processing is complete, the solution was filtered through a porous glass filter and determined the amount of the obtained extract. For the obtained extracts were determined by the content of dry substances of refraction and gravimetric methods and foaming ability.

Optimal conditions for extraction was determined experimentally by the content of dry substances in the extract and its foam-making abilities. The increase in the solids content and foam-making abilities allows to judge about more complete extraction of saponins from the roots Saponaria officanalis L.

The results of experiments on the optimal choice of the extraction conditions are presented in the examples.

Example 1

They took a portion of the roots Saponaria officinalis L. the mass of 6 g, placed in the chemical glass, poured 30 ml of distilled water (water duty 1:5). Leave to soak for 20 minutes. After that conducted ultrasonic treatment was performed using the device IKASONIC U50 control (30 kHz), for which the nozzle of the ultrasonic generator immersed in chemical glass of pre-soaked with raw materials. Ultrasonic action lasted for 20 minutes with intensity 230 W/cm 2 . After extraction, the solution was filtered through a glass filter. The obtained extracts were determined by the content of dry matter and foaming ability.

Under these conditions it foaming ability of the extract was equal 379%, the content of dry substances of 14%.

Example 2

The experience was similar to example 1, except that the process was carried out at the intensity of ultrasonic influence 184 W/cm 2 .

While foaming ability extract was equal to 200%, the content of dry substances of 7.8%

Therefore, the reduction of intensity of ultrasonic influence leads to deterioration of quality of the obtained extracts.

Example 3

The experience was similar to example 1, except that the process was carried out at the intensity of ultrasonic influence 276 W/cm 2 .

While foaming ability of the extract was equal to 43%, the content of dry substances of 8.3%.

Therefore, the increased intensity of ultrasonic influence leads to deterioration of quality of the obtained extracts.

Example 4

The experience was similar to example 1, except that the process was carried out for 15 minutes.

While foaming ability of the extract was equal 346%, the content of dry substances of 13.3%.

Therefore, reduction of time of ultrasonic influence leads to deterioration of quality of the obtained extracts.

Example 5

The experience was similar to example 1, except that the process was carried out for 25 minutes.

While foaming ability of the extract was $ 347%, the content of dry substances of 14%.

Consequently, the increase of time of ultrasonic influence leads to deterioration of quality of the obtained extracts.

These examples show that the optimal conditions for obtaining capodistrias extracts are: ultrasonic treatment intensity 228% to 232 W/cm 2 for 19% to 21 minutes.

The invention is at the stage of laboratory research.
