The invention relates to medicine, namely to the dentist, and can be used to study the etiology and pathogenesis of periodontitis.
There is a method of modeling periodontitis [1], according to which experimental rats injected ammonium chloride at a dose of 3.5 to 4.5 mg per 1 g of body weight once a day for at least 30 days. The disadvantage of this method is that periodontitis is modeled metabolic acidosis, excluding the factors of stress syndrome, which gives incomplete picture of the simulated pathology. In addition, the modeling process very time-consuming, as it requires daily injections in food strictly metered quantity of ammonium chloride.
The closest in technical essence is a method of obtaining a model of periodontitis [2], which is chosen for the prototype.
The prototype method is as follows. Produce the effect of emotional stress on experimental animal by placing 10-11 animals in a cage with square 0,018 m2/individual. The disease develops after a month of exposure.
The disadvantage of the prototype is that the disease is reproduced only when the significant impact of emotional stress on the experimental animal, the elements lesions not typical in their morphological properties for which aragonite in humans. In addition, in animals developing comorbidities characteristic of the General stress syndrome. This reduces the reliability of the simulated pathology. In addition, the modeling process is long because it lasts at least a month.
The task, which is aimed by the invention, is to increase the reliability modeling by obtaining isolated lesions in laboratory animals, is similar to the pathological manifestations of periodontitis in humans, as well as reducing the time for obtaining the model of periodontitis.
The problem is solved in that in the method of obtaining a model of periodontitis by the impact of emotional stress on experimental animals, for example, adult rats, due to space 10-11 animals in a cage with square 0,018 m2/individual, unlike the prototype before placing in the cage each experimental animals under ether anesthesia create an artificial dental plaque around the neck of the upper and lower incisor teeth using dental cement, and dental plaque should have a rough surface and go to 1-1,5 mm under the gums. In the process of modeling the experimental animals get pasty food.
Thus, in the present method of obtaining a model of periodontitis used by the combined effects of local and General the (endogenous) factors on the background of changes in the reactivity of the organism, that corresponds to the modern view of the nature of periodontitis [3].
Example 1: Took 11 individuals Mature rats. Under ether anesthesia was created by dental cement artificial tooth plaque around the neck of the upper and lower incisors with a rough surface. Then the animals were placed in a common cell with square 0,018 m2on the individual. During the whole time of the simulation, the animals received pasty food, excluding the self-cleaning of the teeth. The signs of inflammation of the gums (gingivitis) in the experimental group were observed on the 4th-5th day of the experiment. In its pathological manifestations modeled in animals, the lesion resembles a picture of periodontitis in humans and reproduced with a high degree of reliability.
The proposed method allows to study the etiology and pathogenesis of periodontitis, which is required for the development of preventive measures and treatment of said pathology.
