The technical field

The invention relates to nanotechnology, namely to obtain mineral silicon water (ΜV)containing silicon nanoparticles of amorphous silica (SiO2) and sodium chloride intended for use in medical purposes, in particular, to improve the energy status of the organism, as an immunomodulator, to improve metabolic processes in the body, inactivator toxic proteins of different origin, the treatment of benign tumors, as onkoprotektor, and diabetes 2.

The level of technology

It is known that no living organism can exist without silica. Medicinal properties of silicon have long been known in folk medicine and now he is firmly embedded in clinical practice official medicine. New drug effective therapy (Chuiko OO, Pogorely VK, Pentyuk OO // Medical chemistry and clinical application of silicon dioxide // /1/, is, for example, pelletized powder nanoparticles of fumed silica. He got the name "Silex" and has been used successfully in the treatment of gastrointestinal diseases as the sorbent and the antidote, and as proof of many pathological conditions. Amorphous silica is non-toxic chemically inert material, its therapeutic effect is due except for the sustained fashion with the surface property. Nanoparticles of amorphous silica in solution is capable, as in the case of silica, to bind microorganisms (effect of agglutinative) and inactivate toxic proteins.

However, obtaining nanoparticles of silica in powder form usually pyrogenic method leads to aggregation of individual particles having a size of about 4-5 nm to aggregates with an average size of about 40 nm. This unit size is a barrier to penetration into cells and intracellular actions of nanoparticles, which dramatically reduces the possibility of their therapeutic application.

It is known ("Method for obtaining nanoparticles of silica", patent No. 2426692, authors: Kalinin, A.I., Cardaminetea VV) /2/that certain non-consolidation of particles of SiO2located in the solution can easily penetrate through the plasma membrane of cells without damage to its surface to participate in the intracellular biochemical processes as a surface catalyst for biochemical reactions.

Also known is a method of obtaining silica nanoparticles in aqueous solution (application for invention No. 2012120733 from 18.05.2012, the decision to grant a patent 06.05.2013) /3/, which is more applicable for the solution of biotechnological tasks and to a lesser extent for medical purposes for use in the process of ion exchange resins.

The task of the invention is to develop a method of obtaining a stable solution of silica nanoparticles with a size of 4-5 nm in physiological NaCl solution for immediate use in therapeutic purposes in the form of mineral silicon water (ΜV), therapeutic effect is based mainly on the restoration of the energy functions of mitochondria and, as a consequence, activation of the immune system.

Disclosure of inventions

The invention consists in that the known method of obtaining silica nano Sol of silica by hydrolysis of tetraethoxysilane (TEOS) in a mixture of TEOS : ethanol : water (mol.) acidified with HCl to a pH of 1.5 to 2=1:5:6 followed by exposure of the mixture at a temperature of 40-50°C for two hours using ion-exchange resin, and perform dilutions of silica nano Sol physiological NaCl solution.

- According to the claimed invention, for a more complete reaction of the hydrolysis of TEOS maturation of silica nano Sol is carried out at a temperature of 55-65°C for 1.5 hours.

- According to the claimed invention, in the process of maturation of silica nano Sol to remove ethanol spend evaporation of the mixture with the reduction of the volume by 1/3.

- According to the claimed invention, to obtain ΜV spend dilution of silica nano Sol heated to 55-65°With physiological NaCl solution, heated to 40-50°C in two stages in equal amounts.

- According to the claimed invention, the ratio of the volume of the source of silica nano Sol: physiological NaCl solution is 1:14.

- According to the claimed invention, after each dilution of silica nano Sol saline solution stand for 15 minutes at 55-5°C.

- Technical result is the following:

- Get clear, colorless mineral silicon water (ΜV) with weight concentration of SiO2nanoparticles of 0.9-1% NaCl and 0.8-0.9 percent and a pH of 6-6 .5.

- Get a MAC, in which the silica is resistant to coagulation due to the buffer action of NaCl.

- Get a MAC, the expiry date which is 2.5-3 months.

- Get a MAC with high biological activity and medicinal properties, caused mainly by the catalytic action of the nanoparticles in the mitochondria of cells, increasing energy efficiency of mitochondria, as well as the inactivation of toxic proteins.

The rationale put signs

First get ΜV containing silicon nanoparticles of silica SiO2the size of 4-5 nm, using sour nanosol silica obtained by hydrolysis of TEOS in ethanol and aqueous HCl with subsequent maturation of the nanoparticles at a temperature of 55-65°C and evaporation of ethanol by reduction of the volume of the mixture by 1/3. Processing of silica nano Sol diluted anion-exchange resin to remove the acidity is eliminated, and the tolerable pH of 6-6 .5 is achieved by dilution of silica nano Sol is heated to 40-50°With physiological NaCl solution in two stages with delay after each dilution for 15 minutes at a temperature 55-6°C with getting MACS resistant to coagulation of silica.

Despite the fact that the weight concentration of silica in the produced mineral water of 0.9-1%, its biological activity (defined A.A. Milov method "Method of control of biologically active drugs ' patent No. 2398227) /4/ is extremely high, and when using MACS for medical purposes requires additional dilution in the range of 5-15 drops ERM 50 ml of water depending on the observed pathology, condition and age of the person. The number of nanoparticles in the treatment portion ΜV remains high in the order of 1012particles. Freely penetrating through the cell membrane, nanoparticles SiO2then get into the mitochondria of cells, where they are, corresponding to the size and function of the ribosomes of mitochondria, involved in the oxidative decomposition of organic acids, as an additional surface, acting as a catalyst bioenergetic processes.

An example of the method

Nanosol silica prepared from a mixture of reagents: TEOS : water, acidified with HCl to a pH of 1.5-2 : ethanol taken in a molar relationship of 1:5:6 respectively. For nanoparticles of silica with a size of 4-5 nm thoroughly mixed the mixture of reagents is kept in an open vessel at a temperature of 55-65°C for 1.5 hours. The volume of the mixture decreases due to evaporation of the ethanol, n is 1/3. Then nanosol diluted with saline solution (0.9% NaCl in distilled water), heated to 40-50°C) in a ratio of 1:7, stirred and maintained at a temperature of 55-65°C for 15 minutes. Then perform the second dilution is also a physiological solution at a temperature of 40-50°C, to a final volume ratio of nanosol : MAC 1:15 and again maintained at a temperature of 55-65°C for 15 minutes. Then, the obtained UV cooled to room temperature and poured into a transparent colorless glass containers for storage and use. ΜV obtained in this way, resistant to clumping of individual nanoparticles and coagulation of silica for 2.5-3 months.

The method of obtaining mineral silicon water using silica nano Sol of silica obtained by hydrolysis of tetraethoxysilane in a mixture of TEOS : ethanol : water, acidified with Hcl being 1:5:6, characterized in that nanosol get at a temperature of 55-65°C for 1.5 hours with the evaporation of ethanol to reduce by 1/3, then spend the dilution of silica nano Sol obtained physiological NaCl solution in 2 stages equal portions saline, pre-heated to 40-50°, the ratio of the volume of the original nanosol : saline 1:7 with an interval of 15 minutes, and after each dilution tempera is ur the solution is kept in the range of 55-65°C.
