An important component of complex therapy of patients with diseases of the hepatobiliary system are hepatoprotectors, which aims to normalize the metabolism, functions and structure of the liver parenchyma. Currently, for the treatment of the aforementioned diseases along with such synthetic drugs as "Heptral and preparations containing essential phospholipids ("Essentiale", "Phosphogliv"), are widely used preparations from plant materials ("Legalon", "Kars", "Liv-52"), based on the mechanism of action of which is an antioxidant and choleretic activity (Matveev A.V., konyayeva H., Kurchenko VP, schekatikhin A.S. Based on the above and the fact that most plant hepatoprotectors are products of foreign manufacture, the development of new drugs of plant origin for the treatment and prevention of liver disease on the basis of raw materials grown on the territory of Russia, is one of the urgent tasks of pharmacy and medicine.

The disadvantages of this method are:

- inability to obtain a dry, free-flowing, stable product that can be used in the production of other dosage forms such as capsules, tablets.

The technical result of the claimed invention is the expansion of the range of plant hepatic and creating a more stable form of the extraction of the drug (dry extract), hepatoprotective actions of columns with stigmas of maize, which contains no pharmacologically maindifference solvent (ethyl alcohol) and convenient for subsequent fabrication based metered-dose oral solid dosage forms.

The technical result is achieved by the fact that the columns with stigmas of maize by izmelchennostju 3-5 mm is extracted with ethyl alcohol 60% at room temperature and the ratio of raw material: extractant - 1:5.5 method of multistage countercurrent extraction with a complete cycle of the battery 5 diffusers at the time of extraction at each stage of extraction 4 hours, additionally extracted the spent raw materials 4 and 5 the diffusers used by the extractant, etc the time of extraction 4 diffuser for 4 hours, then drained and passed through 5 diffuser, soaking for 16 hours, then wring out the waste of raw materials, then the combined extract advocate for at least 3 days at a temperature of 2-10°C, the purified extract concentrated under vacuum at 40-50°C, dried in a vacuum oven at 40-50°C to obtain dry extract and crushed.

The authors suggest a way to get funds that have hepatoprotective effect, to expand the range of drugs hepatoprotective activity and create a more technologically advanced form of extraction of the drug (dry extract) from this raw material.

The optimal ratio of phases raw material extractant for columns with stigmas of maize with izmelchennostju 3-5 mm is 1:5,5, because it provides the necessary amount of "mirror" of the extractant (free extractant layer) over the surface of the raw material selected additives.

M.: Medicine, 1976, 202 S.) the four most commonly used in the pharmaceutical industry methods of extraction: 2 and 3-speed ramezanali, multistage countercurrent extraction (IPE) with a complete cycle in a battery of 5 or 6 diffusers. The data obtained indicate that the highest yield of biologically active substances can be achieved with 3 speed ramezanali, but it is necessary to use a considerable amount of extractant per 100 g of material, which can further lead to increased energy and economic costs of production, because at the final stage of obtaining a dry extract will need to remove significant amounts of extractant. Theoretical output of BAS when using a multi-stage countercurrent extraction with a complete cycle of the battery 5 diffusers are somewhat lower, but it involves the reduction and the use of extractive agent per 100 g of the raw material is almost 2 times (compared with rematurely) and more concentrated extracts. Increasing the number of diffusers to 6 impractical, since the increase of the yield of biologically active substances is negligible and accounts for 1.3% (Fig.9).

To extract corn columns with stigmas were studied following variants of the above method: extraction in the equilibrium conditions (duration-stage extraction - 4 hours) and in non-equilibrium conditions (duration of stages 3 hours). The extract yield obtained in the equilibrium conditions, flavonoids and phenolic acids by 24% and 12.5%, respectively, higher than the same indicator of the extract obtained in non-equilibrium conditions (figure 10).

The implementation of this method in practice confirmed data R. the accounts (figure 10). The flavonoids and phenolic acids increased almost by 20% and amounted to 88% and 85,46%, respectively.

The obtained extract is thoroughly mixed and produced purification by sedimentation at a temperature of 2-10°C for at least 3 days before the formation of a dense precipitate, and then filtered.

The purified extract is condensed under vacuum at 40-50°C, and then dried in a vacuum drying Cabinet at a temperature of 40-50°C to obtain dry extract data regimes prevent the degradation of BAS, as evidenced by the high output flavonoids and phenolic acids at the stage of drying (11) and further stability of the active ingredients of the extract during storage (Fig). Conducted research to determine the hepatoprotective activity of the extract of corn columns with stigmas dry in doses of 100 and 200 mg/kg on a model of acute toxic hepatitis caused by the introduction of carbon tetrachloride, 59 white Wistar rats.

To study the effect of corn extract columns with stigmas dry on the course of acute hepatitis white Wistar rats of both sexes (weighing 150-200 g) were divided into 5 groups of 10-15 animals. The animals were kept in standard vivarium conditions on a standard diet and free access to food and water. Experimental acute hepatitis caused by intragastric administration to rats of 1 ml of 25% oil solution of carbon tetrachloride for 6 days (group of animals No. 2-5), intact animals (group # 1) received peach oil. Hepatoprotective effect of the preparations was studied by oral administration once a day for 21 days in a therapeutic mode, i.e. after the formation of the model of pathology. The first and second groups (intact control animals, respectively) was given via a stomach tube, 0.5 ml of starch paste 2%; the third group - aquioben quantity of starch paste containing corn extract columns with stigmas dry (100 mg/kg); the third corn extract columns with stigmas dry (200 mg/kg); the fourth - drug comparison Legalon (200 mg/kg).

The resulting experiment data were subjected to statistical analysis. Data are presented as M±M, where M is the mean value, m is the standard error of the mean. The samples according to the law of normal distribution was assessed by the criterion Shapiro-Wilke. To test statistical hypotheses about the difference between the study groups in the case of abnormal distribution used nonparametric test Mann-Whitney, in the case of normal distribution - parametric criterion of student. Changes of biochemical parameters of blood of the animals of the control group was assessed by comparison with the same indicators in intact animals; group receiving the experimental drug in different doses and medication comparison to the control animals.

Intoxication by carbon tetrachloride was accompanied by hyperferritinemia, indicating the destruction of the hepatocytes and the development of cholestasis (Fig). In the treated group, corn extract columns with stigmas dry in the dosage of 100 mg/kg, there was a statistically significant reduction in the activity of ALT, GGT and alkaline phosphatase, comparable to prep ratom comparison. The introduction of carbon tetrachloride was characterized by disorders of all kinds of currency and was accompanied by an increase in the serum of control animals cholesterol, triglycerides, urea and bilirubin, as well as reduced concentrations of glucose and albumin (Fig). Administration to rats of corn extract columns with stigmas in the dosage of 100 mg/kg resulted in reduced levels of cholesterol, triglycerides and urea in blood plasma compared to the control group. In rats treated with the extract of corn columns with stigmas dry in the dosage of 200 mg/kg, was partially restored glucose level. In all cases, the effect of the experimental drug was comparable or exceeded the effect of legalon.

Thus, the results of the experiments revealed that the introduction of experimental animals with corn extract columns with stigmas dry in the dosage of 100 mg/kg accompanied by a decrease giperfermentemii and restoration of the main indicators of lipid and carbohydrate metabolism and the synthesis of urea.

An example implementation of the method

The content of total flavonoids in the extract is 1,71±0,03%, phenolic acids - to 4.81±0,15%, the loss in weight during drying of 3.46±0,15%.

The inventive method of receiving means having a hepatoprotective effect, has the following advantages over known: lack of end product maindifference for the body of the extractant (ethyl alcohol) and create a more stable form of the extraction of the drug (dry extract) for further use in the production of other technologically more advanced dosage forms. Identified in a model of acute toxic hepatitis activity allows you to create a drug hepatoprotective actions for use in therapy, in particular, in gastroenterology.
