1,2-disubstituted heterocyclic compounds

FIELD: chemistry.

SUBSTANCE: described are 1,2-disubstituted heterocyclic compounds of formula (I) where HET, X, Y and Z values are presented in description, which are phosphodiesterase 10 inhibitors. Also described are pharmaceutical composition and methods of treating central nervous system (CNS) disorders and other disorders, which can influence CNS function.

EFFECT: among disorders that can be subjected to treatment, there are neurological, neurodegenerative and psychiatric disorders, which include, but are not limited by them, disorders, associated with impairment of cognitive ability or schizophrenic symptoms.

2. The compound according to claim 1, where Z represents optionally substituted heteroaryl containing only 6 atoms in the ring, or optionally substituted heterobicyclic ring system.

Connect the imposition of any one of claims 1 to 2, where Z represents an optionally substituted heteroaryl containing only 6 atoms in the ring selected from C and N, provided that the total number of nitrogen atoms in the ring is less than or equal to two; the specified ring optionally is not more than 2 substituents independently selected from C1-C4alkoxy and halogen.

4. The compound according to any one of claims 1 to 2, where Z is benzimidazolyl, chinoline, tetrahydropyranyl, imidazo[1,2-a]pyridine-2-yl, tetrahydroisoquinoline, 5-methylpiperid-2-yl, 3,5-dimethylpyridin-2-yl, 6-verginelli or ethanolic, each of which is not substituted or is substituted by not more than 3 substituents, independently selected from the group consisting of C1-C4alkoxy and halogen.

5. The compound according to claim 1, where X represents phenyl, substituted phenyl or pyridyl.

The compound according to claim 1, where X represents a benzo[d]oxazolyl, benzo[C][1,2,5]oxadiazolyl, benzo[C][1,2,5]thiadiazolyl, benzo[d]isoxazolyl, 1H-benzo[d]imidazolyl, benzo[d]thiazol, benzo[C]isothiazole, benzo[d]isothiazole, benzo[C]isoxazolyl, imidazo[1,2-a]pyridinyl or imidazo [1,5-a]pyridinyl.

11. Pharmaceutical composition having inhibitory activity against phosphodiesterase 10 containing a therapeutically effective amount of the compound or its pharmaceutically acceptable salt according to any preceding paragraph, and a pharmaceutically acceptable carrier or excipient.

A method of treating Central nervous system disorders associated with inhibition of the FOSFA diesterase 10, including the introduction of a person in need, a therapeutically effective amount of the pharmaceutical composition according to claim 11, having inhibitory activity against phosphodiesterase 10, in a suitable dose of 0.01-100 mg per kilogram body weight of recipient per day.

13. A method of treating eating disorders, compulsive attraction to gambling, sexual disorders, including the introduction of a person suffering from such a disorder, a therapeutically effective amount of the pharmaceutical composition according to claim 11, having inhibitory activity against phosphodiesterase 10, in a suitable dose of 0.01-100 mg per kilogram body weight of recipient per day.

14. Method for the treatment of schizophrenia, schizo-affective conditions, disease, Huntington chorea, dystonic States and tardive dyskinesia, including the introduction of a person suffering from such a disorder, a therapeutically effective amount of the pharmaceutical composition according to claim 11, having inhibitory activity against phosphodiesterase 10, in a suitable dose of 0.01-100 mg per kilogram body weight of recipient per day.
