The invention relates to medicine and biology and can be used in the manufacture of histological preparations in the laboratory.

Known framework for the conclusion of histological preparations (Merkulov, G. A. Course histopathological techniques. - M.: Medicine, 1969. - p.132), which used a solution of polystyrene in xylene at the rate of 30 g per 100 g by addition of the plasticizer of dibutylsebacate 6 g per 100 g

The disadvantages of this structure are:

- the duration of the preparation of the basic solution;

- the length of his hardening;

- rapid evaporation and the need to restore the desired consistency;

- solubility in water ranges from 0.005% at 25C, which can lead to deterioration of the quality of the product;

- the presence of 1.6% of water, which can also affect the quality of the product, facilitating its flooding;

Known environment on the basis of polystyrene in xylene, where 25 g is taken 68 g of xylene and 6 g of dibutyl phthalate (www.biooptics.narod.ru; Barshteyn R.S., K. I., J.E. (Plasticizers for polymers. - M, - 1982, s). The disadvantages of this environment are:

- a large number of raw materials;

- a long time for preparation of working solution;

high volatility in such concentrations and the need in this regard, the frequent addition of solvent. In this case violated Rasnov is this between the resin and plasticizer

Known analog - prototype:

environment for the conclusion of histological sections based on the solution of polystyrene in xylene with addition of dimethylphthalate (patent No. 2117273 from 10.08.1998). The disadvantages of the prototype is that

- when heated results in the release of formaldehyde, resulting in discoloration of the product;

- massive use without the cover glass, it is highlighted in the surrounding air;

the necessity of establishing for the storage of drugs to certain conditions;

- the film is easily removed from the slide;

- the relatively high cost from 2500 to 3628 rubles for 1000 ml

- higher density plasticizer;

- does not require use of cover glass, which reduces the stability of the drug to external influences, i.e. objectivity suffers morphological changes in the slice;

next you need special storage conditions: without access to direct sunlight;

is nephrotoxic and neurotoxic.

The invention is aimed at providing cost reduction and casting environment and improving the quality of tissue specimens while maintaining dyeing.

This technical result is achieved by the fact that, as the resin take a polyurethane foam (PUF), solvent - xylene and an auxiliary component dibutyl the ri, the following proportions of ingredients, wt.%

Polyurethane foam15-20
Xylene79-84
Dibutyl1,0

As a result of the studies determined the optimal concentration of ingredients (table 1), ensuring the quality of the drug.

The invention is illustrated in the following examples provide cost reduction and casting environment and improving the quality of tissue specimens while maintaining dyeing.

Example 1. To obtain the composition of the foam, if necessary, crushed and mixed in a ratio of wt.% xylene 84, PPU - 15, dibutyl phthalate and 1.0. The resulting mixture was stirred by shaking. Dissolution occurs within 2-3 minutes. To conclude on a glass slide with colored slice, put a drop of the composition so that it completely covered the cut. Then cover cover glass. Complete drying occurs within 10 minutes. The drug is ready for microscopy.

Example 2. To obtain the composition of the foam, if necessary, crushed and mixed in a ratio of wt.% xylene 81, PUF - 18, dibutyl phthalate and 1.0. The resulting mixture was stirred by shaking. The dissolution occurs in t is the increase of 3-4 minutes. To conclude on a glass slide with colored slice, put a drop of the composition so that it completely covered the cut. Then cover cover glass. Complete drying of the drug comes in 15 minutes. The drug is ready to microcapillary.

Example 3. To obtain the composition of the foam, if necessary, crushed and mixed in a ratio of wt.% xylene 79, PPU - 20, dibutyl phthalate and 1.0. The resulting mixture was stirred by shaking. Dissolution occurs within 3-5 minutes. To conclude on a glass slide with colored slice, put a drop of the composition so that it completely covered the cut. Then cover cover glass. Complete drying occurs within 16 minutes. The drug is ready to microcapillary.

Using our proposed initial components substantially reduces the cost of the environment. The advantages offered by the environment are reflected in table 2.

Thus, the proposed environment in 17.2 in 30,21 times cheaper than the first known environment; 16,32 - 28,73 times cheaper than the second Wednesday in 16,53 -29,21 times cheaper third (table 3). The composition was tested in the preparation of histological preparations in the departments of anatomy, histology, pathological anatomy and pathological physiology.

FIELD: metallurgy.

SUBSTANCE: first, a specimen of rectangular shape is made; a bevelled cut is made on the specimen at an angle of 15-25 from lower base to upper one, thus taking specimen length as the base. Then, surface of the bevelled cut of the specimen is ground and an austenisation mode is performed in oxidation atmosphere by etching gas. The specimen is cooled in water or in the air; then, a microslice or a series of microslices is prepared on surface of small base of the specimen, thus grinding the layers parallel to large base of the specimen. Depth of decoration zone is measured by etching gas on microslice by means of a microscope; then, the investigated surface of microslice is etched with alcohol solution of nitric acid till boundaries of austenitic grains are determined; determined boundaries of austenitic grains are studied; depth of selective determination zone of boundaries of austenitic grains is determined, and the defined etching picture is photographed. As per investigation results of microslice surface, structural state of the specimen is evaluated step by step: first, decoration zone of structure by oxidation with etching gas, then, selective etching zone of actual boundaries of austenite grains, and finally, zone of simultaneous detection of boundaries and intra-grain structure of the investigated steel; then, full depth of penetration of etching gas to the investigated material is determined by summing depths of decoration zone with etching gas and zone of selective detection of boundaries of austenite grains at etching of microslice and multiplication of the obtained value by cosine of inclination angle of bevelled cut to the large base.

EFFECT: simpler detection of boundaries of actual austenite grain; providing complex evaluation of structural state of hardened steel with possibility of multiple layer-by-layer investigation of slices by simultaneous fixation of oxidation zone of the investigated steel, zone of selective detection of boundaries of actual austenite grain and zone of intra-grain structure on slice surface.
