Method for detecting decreased phosphatidyl ethanolamine content in red-cell membranes of pregnant woman suffered aggravated herpes viral infection with underlying increasing peripheral blood tnfα

SUBSTANCE: invention describes a method of detecting the decreased phosphatidyl ethanolamine content in red-cell membranes in a pregnant woman suffered aggravated herpes-virus infection in their third trimester of pregnancy with underlying increasing peripheral blood TNFα, characterised by the fact that peripheral blood of the pregnant woman is analysed for the TNFα content to detect phosphatidyl ethanolamine in red-cell membranes by two-dimensional thin-layer chromatography, and a discriminant equation is presented. Df=+1.743×TNFα+(-1.079×phosphatidyl ethanolamine), and if Df is more than a boundary value 78.86, the decreased phosphatidyl ethanolamine content in red-cell membranes is detected.

EFFECT: higher red-cell membrane stability in peripheral blood of the pregnant woman.

The purpose of the study is to show that during exacerbation of herpes virus infection during pregnancy is increased in the peripheral blood content of TNFα, which decreases the percentage in erythrocyte membranes pregnant phosphatidylethanolamine.

The phospholipids of erythrocyte membranes ensure the integrity of the morphological structure of the erythrocyte and firmly held erythrocyte membrane. The effects on membrane interleukin provides change their viscoelasticity properties, which entails the stability of the membranes of red blood cells.

One of the probable causes of changes in the number of phosphatidylethanolamine is its transformation into phosphatidylcholine by methylation [1, 2, 5, 6] with an increase in the titer of antibodies to the herpes virus.

The prototype for this study we chose the material covering the change in the content of phosphatidylethanolamine in erythrocyte membranes in patients with chronic bronchitis (Lutsenko M.L. Phospholipids in violation of the respiratory functions of the body. Blagoveschensk, Russian Academy of medical Sciences, 2006. - 163 C.) [5]. The author noted that in chronic bronchitis, the content of phosphatidylethanolamine in erythrocyte membranes is reduced by 8.1%. In chronic bronchitis, the number of phosphatidylethanolamine in erythrocyte membranes is 16,16±0,41% (control - 20,17±0,27%). However, unlike e the th work in our study we note, that the decrease of phosphatidylethanolamine in the membranes of red blood cells occurs in pregnant women during exacerbation of herpes viral infection, which appears in the blood of a large number of TNFα. These factors become damaging the phospholipid structure of erythrocyte membranes. Such works available to us in the literature was not found.

During exacerbation of herpes viral infection with an increase in the titer of antibodies to the herpes virus and the increase in blood TNFα number of phosphatidylethanolamine in erythrocyte membranes pregnant reduced.

1. When the titer of antibodies to the G1up to 1:800 content TNFα was 62±1,1 PG/ml;

2. When the titer of antibodies to the G1to 1:3200 content TNFα was 60,0±2.3 PG/ml;

3. When the titer of antibodies to the G1to 1:6400 content TNFα was 166,0±3,2 PG/ml;

4. When the titer of antibodies to the G1to 1:12800 content TNFα was 590,0±20,0 PG/ml [5].

However, the studies did not address the issue, which of the phospholipids of erythrocyte membranes strongest damaging influence TNFα.

The analysis showed that during exacerbation of herpes virus infection in the third trimester of pregnancy when the titer of antibodies to herpes virus 1:12800 number of phosphatidylethanolamine was decreased to 20.3±0,80% (control - 24,1±1,0%) with an increase at this time TNFα to 590,0±20,0 PG/ml (control - 35,7±1,8 PG/ml). Ff(144,00) more boundary values (78,86), therefore, increasing the differential functions of the above boundary values increase the content of TNFα leads to a decrease in the content of phosphatidylethanolamine in erythrocyte membranes against the exacerbation of herpes viral infections.

The claimed method has the following methods:

1. Investigated peripheral blood of 20 pregnant women who have had herpes viral infection in the third trimester of gestation. Control was the study of peripheral blood of 20 pregnant, not ill during the whole period of gestation. Research was conducted on the basis of hospital obstetric clinic of the far Eastern scientific center of physiology and pathology of respiration SB RAMS institutions of the Russian Academy of medical Sciences. All studies were conducted with regard to the requirements of the Helsinki Declaration of the world Association "Ethical principles for medical research involving human subjects" with amendments of 2000, and the "Rules of clinical practice in the Russian Federation", approved by Order of Ministry of health of the Russian Federation dated 19.06.2003, No. 266.

2. The titer of antibodies to the herpes virus was determined by ELISA n the spectrophotometer Stat Fax-2100 (USA).

3. Phospholipids in erythrocyte membranes was determined by two-dimensional thin-layer chromatography [4].

4. TNFα was determined by ELISA method using sets of LLC "Cytokine" (St. Petersburg) on spectrophotometer "Stat Fax-2100" (USA).
