Method of producing 1,7-bis[4(5)-methyl-1,3-thiazol-2-yl]-3,5-dithia-1,7-diazaheptanes

FIELD: chemistry.

SUBSTANCE: invention relates to a method of producing 1,7-bis[4(5)-methyl-1,3-thiazol-2-yl]-3,5-dithia-1,7-diazaheptanes of general formula (1), which can be used in organic synthesis to produce macro-heterocyclic rings, as well as sorbents and extraction agents for precious and rare-earth metals. The method is realised by reacting hydrogen sulphide-saturated aqueous formaldehyde solution (37%) with 2-amino-4(5)-methylthiazole with molar ratio of initial reagents 2-amino-4(5)-methylthiazole: formaldehyde: hydrogen sulphide equal to 20:30:20, at temperature -5-5°C and atmospheric pressure for 8-12 hours.

EFFECT: improved method.

 

The invention relates to the field of organic chemistry, specifically, the method of obtaining 1,7-bis[4(5)-methyl-1,3-thiazol-2-yl] - for 3,5-dithia-1,7-vasagatan General formula (1):

N,N-hetaryl-3,5-dithia-1,7-vasagatan can find application in organic synthesis for the preparation of macroheterocycles, as well as effective sorbents and extractants precious and rare earth metals [Deutsche Gold - und Silber-Scheideanstalt vormals Roessler. F.R. 1341792, 1963 (Chem. Abs. 1964, 60, 5528d)].

The known method (Aleev R.S., galinova US, Popov, Y., Makhutov P.M., Rafikov CF. DAN SSSR, 1988, 4, 873-875) obtain oligomeric amidosulfuron (2) reaction timestribune the research with formaldehyde and hydrogen sulfide according to the scheme:

The known method cannot be obtained 1,7-bis[4(5)-methyl-1,3-thiazol-2-yl] - for 3,5-dithia-1,7-vasagatan General formula (1).

The known method (Binz A.N., Reinhart F.E., Winter NS J. Amer. Soc, 1939. P.3134) obtain 1,7-di(4-morpholine)-2,4,6-tritiated formula (3) reaction of the research with formaldehyde and hydrogen sulfide in the presence of HCl according to the scheme:

The known method does not allow 1,7-bis[4(5)-methyl-1,3-thiazol-2-yl] - for 3,5-dithia-1,7-vasagatan General formula (1).

Thus, the literature contains no information on the selective receipt of 1,7-bis[4(5)-methyl-1,3-thiazol-2-yl] - for 3,5-dithia-1,7-deathgate what s the General formula (1).

We propose a new method of obtaining 1,7-bis[4(5)-methyl-1,3-thiazol-2-yl] - for 3,5-dithia-1,7-vasagatan General formula (1).

The method consists in pre-saturated aqueous solution (37%) formaldehyde hydrogen sulfide followed by the addition of alcohol (96% ethanol) solution of 2-amino-4(5)-methylthiazole taken in a molar ratio aminomethylphenol:CH2O:H2S = 20:30:20. The mixture is stirred for 8-12 hours at a temperature of-5-5°C and atmospheric pressure, produce 1,7-bis[4(5)-methyl-1,3-thiazol-2-yl] - for 3,5-dithia-1,7-vasagatan formula (1) with output 61-72%. The reaction proceeds according to the scheme:

1,7-Bis[4(5)-methyl-1,3-thiazol-2-yl] - for 3,5-dithia-1,7-vasagatan (1) are formed only with the participation of 2-amino-4(5)-methylthiazole, formaldehyde and hydrogen sulfide, taken in a molar ratio of 20:30:20. When a different ratio of initial reagents decreases the selectivity of the reaction. In the presence of other aldehydes (e.g., alkyl-, aryl-substituted aldehydes), other serosoderjaschei compounds (for example, carbon disulfide, diallylsulfide, mercaptans or other substituted thiazolo (for example, Aristotle) target products (1) are not formed.

The reaction was carried out at a temperature of-5-5°C. At temperatures above-5-5°C (40°C) decreases the selectivity of the reaction and increase energy costs, and at temperatures below-5-5°C (for example, -15°C) reduced Corot reaction.

Significant differences of the proposed method.

In the known method the reaction proceeds with participation as a source of compounds of the research, formaldehyde and H2S in the presence of HCl. The known method does not allow 1,7-bis[4(5)-methyl-1,3-thiazol-2-yl] - for 3,5-dithia-1,7-vasagatan formula (1).

In the proposed method, as the initial reagents used 2-amino-4(5)-methylthiazole, formaldehyde and hydrogen sulfide, taken in a molar ratio of 20:30:20, the reaction proceeds in water-ethanol medium without HCl. In contrast to the known proposed method allows you to obtain an individual 1,7-bis[4(5)-methyl-1,3-thiazol-2-yl] - for 3,5-dithia-1,7-vasagatan formula (1).

The method is illustrated by examples.

Example 1. In a glass reactor, equipped with a mechanical stirrer, a gas-feeding tube and addition funnel at 0°C is placed 30 mmol of formaldehyde (37% aqueous solution), saturated with hydrogen sulfide (20 mmol) for 30 min, was added dropwise 20 mmol of 2-amino-4-methylthiazole in alcoholic medium (96% ethanol). The reaction mixture is stirred at a temperature of ~0°C for 10 h, extracted with chloroform, produce 1,7-bis-(4-methyl-1,3-thiazol-2-yl) - for 3,5-dithia-1,7-vasagatan (1) yield 68%. Other examples of the method are given in table 1.
