SUBSTANCE: invention refers to medicine, namely to physiology and physiopathology. Substance of the invention consists in an application of peptide Gly-His-Lys of formula (NH2) Gly-His-Lys (COOH) for an analgesic effect in pain caused by thermal irritation.

EFFECT: invention allows applying known peptide Gly-His-Lys with manifested analgesic effect for a new purpose.

In the literature, many authors have described the properties of the peptide Gly-His-Lys. You know its modulating influence in the regulation of growth and differentiation of cells (Kato, K., Kasai, S., Onodera K. et al. Several studies have confirmed the influence of the peptide as a promoter of wound healing, as is known for its stimulating effect on collagen synthesis by fibroblasts (F.X. Maquart, Bellon G., Chaqour Century. et al. In vivo stimulation of connective tissue accumulation by the tripeptide-copper complex glycyl-L-histidyl-L-lysine-Cu2+ in rat experimental wounds. // J. It is shown that Gly-His-Lys stimulates the physiological and reparative regeneration of the liver (Smakhtin M., Chvanov I.A., Hemp A.A., Babintsev I.I. Object of the invention is a new usage of the peptide Gly-His-Lys, having the formula (NH2) Gly-His-Lys (COOH), analgesic effect for pain caused by temperature irritation.

The objective of the invention is achieved through the use of the peptide Gly-His-Lys for analgesic effect for pain caused by temperature irritation. At the present time, because the known exact chemical formula of the peptide, it can be synthesized in any laboratory synthesis of peptides or purchased from companies involved in the delivery of biologically active compounds (Sigma, ICN, Merk, Bachim and others). We used the drug, synthesized in the Institute of chemistry, St. Petersburg state University. Dissolved in isotonic sodium chloride peptide Gly-His-Lys introduce experimental animals (mice) parenteral (intraperitoneal) once in doses of 0.5 and 5 mg/kg of body weight for 12 minutes before painful irritation.

An example of a specific implementation.

Experimental animals (mouse-male BALB/c) weighing 20-25 g were divided into groups of 10 animals each (table). The animals were kept at the temperature of the air in the room at 22-24°C, light mode: 12 hours light, 12 hours darkness and received article is ndirty pelleted feed and water freely available.

In the experiment, temperature and pain sensitivity can be estimated by the method of "hot plate" (Sarnow L.N., Hazura CENTURIES, Elements of experimental pharmacology. - M., 2000. - 352 C.). For the study of pain sensitivity during thermal exposure, the mice were placed on the plate with the bounding glass cylinder heated to a temperature of 55°C. was Performed 4 tests in 1 min with an interval of 1 min. Pain sensitivity was assessed by the magnitude of the latent periods (in seconds) response licking of the paws in the 2nd test.

The peptide Gly-His-Lys was dissolved in isotonic sodium chloride to obtain the necessary doses of 0.5 and 5 μg/kg animal body weight. Injected parenterally (intraperitoneally) once the experimental animal in volume obtained at a rate of 1 ml per 1 kg of body weight (0.1 ml) for 12 minutes before painful irritation. Control animals received equivalent volumes of saline (FR).

Thus, the results of experimental studies show that the use of the peptide Gly-His-Lys at doses of 0.5 and 5 mg/kg causes a pronounced analgesic effect for pain provoked temperature irritation. This method is based on application of regulatory peptide belonging to the class of biologically active substances with a strong modulatory effect and multifunctional, is not time consuming and does not require sophisticated equipment.
