Method of predicting saturation of peripheral blood of pregnant women with oxygen depending on indices of glucose-6-phosphat-dehydrogenase in erythrocytes with various degree of herpes-virus infection agression and content of tnfα in peripheral blood

FIELD: medicine.

SUBSTANCE: in order to predict insufficiency of saturation with oxygen of peripheral blood of pregnant women who have herpes-virus infection, content of glucose-6-phosphat-dehydrogenase and TNFα in peripheral blood is determined. Discriminant equation D=-0.589·G-6-PDG+{+1.23·TNFα} is solved. If value of discriminant function is equal or greater than 89.95, predicted is development of oxygen insufficiency, accompanied with reduction in peripheral blood of pregnant women of pO2 with titre of antibodies to Herpes simplex virus 1:12800 to 27.63±0.7 mm Hg, and of HbO2 to 90.2±0.47% (control: pO2 - 39.22±0.5 mm Hg; HbO2 - 95.3±0.27%).

EFFECT: increased accuracy of determining possibility of development of oxygen insufficiency in pregnant woman with herpes-virus infection.

The invention relates to medicine, namely to diagnosis.

The aim of the study is the prediction of the degree of saturation in the blood of pregnant oxygen at different aggressiveness of herpes viral infection, depending on the performance of glucose-6-phosphate dehydrogenase in erythrocytes and content of TNFα in peripheral blood.

The prototype was elected estimation method for reducing the content of glucose-6-phosphate dehydrogenase in erythrocytes of peripheral blood when receiving antimalarial drugs (Carson R.E., 1956), changes in the content of glucose-6-phosphate dehydrogenase in erythrocytes of peripheral blood in patients with pneumonia, typhoid fever and influenza (Idelson LI et al., 1973, 1975; Vorob'ev A.I., 2007).

In contrast to the above prototype-the analysis of the dynamics of reduction of glucose-6-phosphate dehydrogenase in erythrocytes of pregnant depending on the titer of antibodies to herpes simplex virus when exposed to TNFα in peripheral blood and are indicators confirming the signs of oxygen deficiency in the peripheral blood during pregnancy on a background of herpes viral infection, that in none of these studies were not performed.

The objective is achieved by the fact that the decrease in the content of glucose-6-phosphate dehydrogenase in erythrocytes of peripheral blood of pregnant women in the period of exacerbation GE is pesnoy infection was carried out by spectrophotometric method using a standard set of "SENTINEL DIAGNOSTICS)) (Italy) on device "Stat-Fax-1900" (USA). At the same time these pregnant women was determined by the content of TNFα using test systems LLC "Cytokine" (St. Petersburg) with a microplate reader (Stat Fax-2100" (USA). Detection of oxygen saturation in the blood of pregnant women was carried out on the analyzer blood gas and electrolytes "CHIRON DIAGNOSTICS-865" (USA). Integral assessment of the relationship of glucose-6-phosphate dehydrogenase in healthy pregnant women (14 cases) and pregnant women who have had an exacerbation of herpes virus infection (14 cases)was performed using the discriminant of the equation by which it was obtained that the most real factor pathological effect on glucose-6-phosphatedehydrogenase was TNFα (step-by-step regression, table 1), and under his influence the content of the enzyme is reduced with 79,4±2,1 IU/l 109red blood cells (control) to 36,39±1,95 IU/l 109red blood cells (titer of antibodies to herpes simplex virus 1:12800). When the titer of antibodies to herpes simplex virus 1:3200 glucose-6-phosphate dehydrogenase was only 48,78±2,3 IU/l 109erythrocytes.

Under the influence of TNFα reduced glucose-6-phosphate dehydrogenase creates a risk of oxygen deficiency in pregnant women with increasing titer of antibodies to herpes simplex virus (1:12800).

Thus, the indicator discriminan the Noah functions is much higher (89,95) boundary values (41,61), reflecting on the influence of TNFα on glucose-6-phosphatedehydrogenase pregnant women with herpes viral infection and reduces the saturation of peripheral oxygen in pregnant women infected with herpes virus (titer of antibodies to herpes simplex virus 1:12800: pO2- 27,63±0.7 mm Hg; HbO2- 90,2±0,47%; the titer of antibodies to herpes simplex virus 1:3200: pO2one - 29.98±0.4 mm Hg; HbO2- to 93.3±0,44%) compared with healthy pregnant women (pO2- to 39.22±0.5 mm Hg; HbO2- to 95.3±0,27%) (table 3).

The technique consists in determination of glutathione reductase and herpes virus antibody titre concentration by the enzyme immunoassay in peripheral blood in pregnant women suffered from a herpes virus infection episode. If glutathione reductase concentration is 8.36±0.137 E/g Hb (ref. - 7.82±0.185 E/g Hb), herpes virus antibody titre is 1:6400. If herpes virus antibody titre is increased to 1:12800, erythrocyte glutathione reductase concentration is decreased 4.48±0.22 E/g Hb.

EFFECT: technique is characterised by high sensitivity and allows predicting the development of impaired respiratory activity of erythrocytes in the pregnant women with the herpes virus infection episode.

SUBSTANCE: simultaneously two parametres are determined: for men - in ejaculate, and for women - in discharge of cervical canal, namely, slgA and lactoferrin. If level of secretory immunoglobulin A is 3.01±0.50 mcg/ml and lower and simultaneous level of lactoferrin is 6876±89 ng/ml and higher, gonococcus infection with systemic manifestations is diagnosed, if said condition is absent, localised gonorrhea is diagnosed.
