Method of diagnosing anthracyclonic cardiomyopathy in experiment

FIELD: medicine.

SUBSTANCE: invention relates to experimental medicine. Bioimpedancemetry (BIM) is carried out three times in the region of the apex of the heart at different points on an area of 5 mm2. The average value (Z avg.), and if the Z avg. is more than 220 Ohm, it speaks of the presence of anthracyclonic cardiomyopathy. The method is simple and makes it possible to reliably diagnose anthracyclonic cardiomyopathy in an experiment.

The invention relates to medicine and can be used in experimental medicine for the detection of anthracycline cardiomyopathy in the experiment.

A known method for the diagnosis of cardiomyopathy in the experiment in which the animal decapitat, performed histological examination of the myocardium. The material is fixed with a 10% solution of neutral formalin, pour in the wax, prepare ultra-thin sections and stained with hematoxylin and eosin. Under the microscope see the signs of cardiomyopathy, such as the presence of disintegration of muscle fibers, sliminate myocytes along the muscle fibers, hyperchromeity nuclei, lipid infiltration of individual cardiomyocytes, a honeycomb structure of the myocardium (Lushnikova EL, Konnikova MG, Young OP, Nepomnyashchikh L.M. Morphological manifestations of cardiac remodeling in dilated cardiomyopathy antratziklinova Genesis. // Bulletin of experimental biology and medicine.- 2004 - volume 138. No. 12, p. 686).

The method is time-consuming, and assessment of cardiomyopathy is subjective.

Object of the invention is objectifying method for the diagnosis of anthracycline cardiomyopathy in the experiment to further study the cardioprotective properties of pharmacological drugs or substances.

The essence of the proposed method is that p is avodat bioimpedance (BIM) of the myocardium at the apex of the heart three times with subsequent calculation of the average value (Zcp.), and if Zcp. 220 Ohm, indicate the presence of anthracycline cardiomyopathy.

Experimentally 10 animals we have determined that the normal value of the impedance of the myocardium of rats is in the range from 175 to 220 Ohms.

Example. Rat weighing 200 g was modeled anthracyclinone cardiomyopathy, which once intraperitoneally injected doxorubicin hydrochloride at a dose of 10 mg/kg

On day 7 after injection of doxorubicin hydrochloride under ether anesthesia produced thoracotomy. Using needle electrodes were BIM (current frequency of 10 kHz, the voltage of 1.02 V) of the myocardium at the apex on the beating heart, which was introduced electrodes of inert metal in the tissue to a depth of 1 mm, the Impedance measured sequentially at different points (Z1, Z2, Z3square 5 mm2. Then calculate the average value (Zcp.).

In the measurement of Z1=230 Ω, Z2=253,25 Ω, Z3=259,49 Ohms, then Zcp.=247,58 Ohms, more than the 220 Ohm, consequently, we can speak about the presence of anthracycline cardiomyopathy.

Next, the animal was decapotable, myocardium were taken for histological examination. By microscopy revealed the presence of signs of cardiomyopathy: evidence of disintegration of muscle fibers, sliminate myocytes along the muscle fibers, hyperchromeity nuclei, lipid infiltration of individual cardiomyocytes, aceito the structure of the myocardium, that confirmed the BIM.

Conducted experimental simulation of anthracycline cardiomyopathy in 10 experimental animals (rats Wistar rats weighing 180-200 g) by the method described above. All animals produced BIM attack, data are tabulated (table 1).

In all rats the values of the impedance of the myocardium of the apex of the heart were more than 220 Ohms, and histological examination revealed signs of anthracycline cardiomyopathy. The average impedance value in animals with anthracycline cardiomyopathy and intact animals, respectively, were equal to 259,5±6.9 Ohms and 198,19±4,16 Ω (p<0,001) (table 2).

Thus, the proposed method for the diagnosis of anthracycline cardiomyopathy in the experiment is simple, objective, reliable, can be widely used in experimental medicine to study the cardioprotective properties of pharmacological drugs or substances.
