Method for stimulating the growth in pseudomonas microorganisms at wheat cultivation

SUBSTANCE: the present innovation deals with stimulating the growth of humus-forming microorganisms at wheat cultivation due to carrying out pre-sowing treatment of wheat seeds by applying solution of a preparation being an aqueous solution of the mixture of a triple copolymer of acrylic acid, its amide and triacryloylhexahydro-1,3,5-triazine (CPAA) and Cubaxin as a growth stimulant at the following ratio of ingredients, weight%: CPAA 0.0005-0.001, Cubaxin 0.0005-0.001, water - the rest, moreover, wheat seeds should be treated due to slurry disinfection technique. The suggested method enables to increase the number of Pseudomonas microorganisms in rhizosphere and rhizoplane of wheat roots.

The invention relates to the field of agriculture, particularly to soil science, and method of promoting the growth gumusoobrazovanie microorganisms for wheat.

Intensification of agricultural production, chemical pollution and environmental degradation has led to depletion of humus horizon of the famous Russian black and progressive degradation processes homeoapathy. Effective ways to stimulate growth of microorganisms in the soil and, in particular, in the rhizosphere and rhizoplane roots of plants does not exist.

It is known that pre-treatment of soybean seeds with an aqueous solution of the potassium salt of 2,4,6-Tris(p-carboxymethylamino)-contrasena (Cubicin) General formula:

Known heterochain ternary copolymer of acrylic acid, its amide, triacilglycerides-1,3,5-triazine (SPACH) as an antistatic agent [Smolevichy, Vinsanity, Linkreplacer, Glu, Suissenautic, Hemogen, Vasagatan. An antistatic composition comprising hexahydrotriazine copolymer. - Vesti Academy of Sciences, 1989, No. 5, s-117]. The use of this copolymer as a stimulator of the growth of microorganisms of the genus Pseudomonas in the cultivation of wheat is generally unknown.

Technical solution to the problem is to stimulate the growth of microorganisms of the genus Pseudomonas in the cultivation of wheat.

The task is achieved in that in the method of intensifying the growth of microorganisms when grown wheat to increase the number of microorganisms of the genus Pseudmonas as a means of using an aqueous solution of a mixture of copolymer what acrylaway acid, it amide, triacilglycerides-1,3,5-triazine (SPACH) and poststimulatory Sebacina in the following ratio of ingredients, wt.%:

while the seeds are treated using dry etching.

The novelty of the claimed proposal is seen that the use of funds on the basis of a mixture of ternary copolymer SPACH and Sebacina for pre-treatment of wheat seeds allows the use of special bacterial drugs to increase the number of microorganisms of the genus Pseudomonas in the rhizosphere and rhizoplane root systems of growing plants. The tool contains a lot of components available and has higher activity in comparison with the prototype application - poststimulation Sebacina.

This triple copolymer SPACH receive and purified in accordance with known methods, by copolymerization of acrylic acid, its amide, triacilglycerides-1,3,5-triazine in an aqueous solution under the influence of redox initiating system at a ratio of initial monomers providing the ultimate chain termination and branching [Smolevichy, Vinsanity, Linkreplacer, Glu, Suissenautic, vs Soldatov. An antistatic composition comprising hexahydrotriazine copolymer. Vesti Academy of Sciences of the BSSR. 1989, No. 5 c.115-117 ]. For comparison, using the prototype application - poststimulatory Cubicin, which is produced and purified in accordance with the patent [see U.S. Pat. Of the Russian Federation No. 2083568. - Salt triazinetrione acids and means for presowing treatment of seeds of agricultural crops on MCL 6 07 D 251/54, a 01 N 43/68].

In the proposed method of preparing a simple tool by dissolving the required quantities SPACH or Suboxine in water at ordinary temperature. The prepared solution is treated wheat seeds varieties Turtledove using dry etching, at the rate of 10 l to 1 ton of seeds. In control variants seeds before sowing treated similarly, with distilled water at the rate of 10 l/tonne of seed. Then the seeds are dried in an air-convection mode, leak filled with soil germinators, containing 400 g of leached Chernozem, slabovolnogo, and grow in the adopted the same for each variety of plant conditions of humidity and temperature. The repetition of experiments is threefold.

On the 7th day the seedlings, together with fragments of soil on the roots are then removed from the growth chamber to determine the number and generic composition of the microflora in the rhizosphere of roots known method [see Methods of soil Microbiology and biochemistry. To do this, the roots of the seedlings together with prelapse the soil, placed in a conical flask, washed by shaking on a shaker for 3 min in sterile water, the resulting suspension filtered through filter paper and the filtrate is subjected to serial dilutions in sterile water. Seeding of microorganisms obtained from dilutions carried out in Petri dishes on solid medium: agar of čapek and MPA and maintained under standard conditions. Counting and identification of bacteria is carried out on 3 days. Determine the average number of colonies in a Cup and carry out the terms of 1 g of absolutely dry soil by the formula: A=b··g/d, where a is the number of colonies in 1 g of soil; b - average number of colonies in the Cup; in - breeding, which is made from the sowing; the number of drops in 1 ml of suspension; D. - weight abs. dry soil. The latter was determined after drying on the filter soil to constant weight at 105°C.

The number of the microflora rhizoplane determined by the method of successive ottavani method Tepper [see Esepa, Vchelyshkov, Heeresarchiv. - Workshop on Microbiology. M: Agropromizdat, 1987. - 239 S.]. After flushing rhizosphere soil the roots consistently placed in 6 flasks containing 100 ml of sterile water, shaken in each of the flasks for 2 min, then the contents of all the flasks are combined and prepare serial dilutions from whose conduct seeding on above the dense medium.

The number of microorganisms in rhizoplane 1 g of roots (A) determined by the formula: A=20A·600V/m, where a is the number of colonies in a Cup, grown from a single planting, taken from breeding a volume of 0.05 ml; in - degree of cultivation; m is the mass of raw roots. The latter is determined by weighing the roots after removing them from the water with filter paper. For definitions of generic and species of microorganisms used in terms of their cultural, morphological and fiziologicheskii properties by a known method [see Jholt, Ntrig, PSNI, Jsteele, Swilliams. Examples of the use of the proposed method of promoting the growth of microorganisms in the rhizosphere and rhizoplane roots of wheat are presented in table (examples 1-5 in table; see the end of the description).

As can be seen from the table data, the application of the proposed method of promoting the growth of microorganisms in wheat (examples 1-5) allows to increase the number of microorganisms of the genus Pseudomonas in rhizoplane and the rhizosphere of the roots of wheat plants on 232,8-251,0 and 145,3 at 149.5% respectively in comparison with control (45,6-48,1 and 145,3 at 149.5 million/g of roots and soil instead of 13.7 and 60.0 m/g of roots and soil, respectively) at that time, as pre-sowing spraying of wheat seeds in an aqueous solution of known means Sebacina increases this item is index only 77.4 and 28.0%, respectively (24,3 and 76.8 m/g of roots and soil).

Thus, the application of the proposed method of promoting the growth of microorganisms by pre-treatment of wheat seeds means on the basis of an aqueous solution of a mixture of ternary copolymer SPACH and poststimulatory Sebacina allows to increase the number of microorganisms of the genus Pseudomonas in the rhizosphere and rhizoplane roots of wheat plants.
