Method for identifying isoniazide

SUBSTANCE: the present innovation deals with identifying isoniazide, pyridine derivative, applied to control the quality of production manufactured by pharmaceutical enterprises and prepared by drug stores. One should detect by treating the sample under investigation with freshly prepared 10%-hydroxylamine hydrochloride alkaline solution at obtaining the color. The method creates minimal duration for detection.

The invention relates to pharmaceutical chemistry, in particular to a method of identification of isoniazid, a derivative of pyridine.

Known methods of identification of isoniazid using 2,4-dinitrochlorobenzene, copper salts or solution of iodine /"a Guide to laboratory exercises in pharmaceutical chemistry. - M., 2001, s-259/. The disadvantages of this method is the relatively low detection sensitivity and obtaining unstable colored reaction products.

Closest to the present invention is a method of identification of isoniazid by treatment with ammonia solution of silver nitrate and then heated in a boiling water bath /State Pharmacopoeia". - M, Medicine, 10th edition, 1968, Istanbul, p. 378/.

The disadvantages of this known method include low sensitivity, poor specificity of the definition, the use of expensive and not readily available reagent silver nitrate, the long duration of the study due to prolonged heating of the reaction mixture in a boiling water bath, and observation of initial fixation of unstable yellow, turning into gray, and then in the selection of metallic silver mirror.

The objective of the invention is to increase the sensitivity, specificity and reducing research time.

The set task is solved by the method namely, that the analyzed sample is treated svezheprigotovlennym 10% of an alkaline solution of hydroxylamine hydrochloride, giving isoniazid yellow staining.

A specific example of the method: to a solution of the investigational product /isoniazid/ add freshly prepared 10% alkaline solution of hydroxylamine hydrochloride, a characteristic staining yellow in color, stable in time. Sensitivity reactions - 1,0·10-4g/ml. the research Time is 3-5 minutes.

The method of the invention in comparison with the selected prototype allows to increase the detection sensitivity 50 times and reduce research time by 5-8 times.

The method of identification of isoniazid by treating the sample with an alkaline reagent of fixing coloring, characterized in that the reagent is used as freshly prepared 10% alkaline solution of hydroxylamine hydrochloride.

SUBSTANCE: method comprises: sampling, acidifying samples with HCl/H2SO4 mixture, adding Ce(SO4)2 as oxidant and removing its excess with reducing agent NH2OH·HCl, adding rhodamine C as organic reagent, extracting resulting complex, separating organic phase from aqueous phase, and measuring optical density.
