Antiviral agent

The invention relates to medicine and refers to the local application of the effect, for example, in dentistry, surgery, otolaryngology, gynecology for the treatment of herpes and other infections caused by viruses, bacteria, namely antiviral agent containing human leukocyte interferon and the substrate in the form of plate, as a basis containing collagen, the activity of interferon in the plate is not less than 3000 ME. Technical result: the creation of a new drug for topical application, containing human leukocyte interferon in the form of a plate having biological availability, and usability. table 1.

The invention relates to medicine and refers to the local application of the effect, for example, in dentistry, surgery (treatment of wounds, burns, trophic ulcers, bedsores), otolaryngology, gynecology for the treatment of herpes and other infections caused by viruses, bacteria. It is known that the most promising antiviral drug is a human leukocyte interferon containing complex helikopterlerin most effective for the prevention and treatment of local lesions, as various eye diseases (herpetic keratoconjunctivitis), skin lesions caused by viruses lesions of the genital organs [3].

Known human interferon - leukocyte dry preparation in ampoules containing not less than 1000 IU of antiviral activity, applied intranasally after dilution with water in the form of drops or in the form of applications wipes, pre-moistened with a solution of interferon [4]. The disadvantage of such dosage forms is the inconvenience of use for the treatment of viral diseases of the mucous membranes and skin, loss of active substance, wetting napkins, tampons, and a small exposure of the drug by intra due to leakage of the solution from the nasal passages.

Known ointment containing recombinant interferon-alpha-2 activity at least 1.7107ME, polyvinyl alcohol, gel, aluminum hydroxide [5]. The tool is designed to treat herpes infections of the skin and mucous membranes. However ointment contains chemicals that can cause allergic reactions. Also preferred therapeutic effect is given to preparations containing human leukocyte interferon, Koskin leukocyte interferon "Intern" for the treatment of viral, bacterial and chlamydial infections, containing as the basis vaseline, surfactant (emulsifier T-2 or monoglycerides) and water [6]. The activity of interferon is 2400-1200 ME. Ointment based on fat in the early stages of wound healing unacceptable.

Known ointment with human leukocyte interferon "Interoir" (prototype) for the treatment of viral diseases, with the activity of interferon 3000 IU/g, containing as the basis of polyethylene oxide and water [7].

Listed tools [6, 7] also contain chemical components, and contact with interferon affected part of the ointment forms less long than the proposed dosage form. In addition, the topical form of the drug is not always preferred for use in gynecology, in the treatment of bedsores, trophic ulcers, prevent the outflow of fluid.

Object of the invention is the creation of new medicines for topical application, containing human leukocyte interferon in the form of a plate having biological availability and ease of use.

As the basis for the proposed dosage form used natural polymer is collagen, which in turn is biological is about the role in the implementation of the reparative function of connective tissue. Collagen preparations may be used as an effective wound dressings [8].

The technical result of the claimed medicinal product: 1) increase therapeutic effect by providing prolonged action of interferon, immobilized on a slowly absorbable sponge; 2) the absence of chemical components; 3) a combination of antiviral action of interferon and wound healing actions of collagen; 4) the sterility of each individual packaging for single use; 5) ease of use in gynecology: not required auxiliary materials (sterile cotton-gauze swabs when applying ointments).

The task is solved in that antiviral topical application, containing the human leukocyte interferon and the substrate in the form of a plate and as the basis contains collagen, the activity of interferon in the plate is not less than 3000 IU.

Created a new antiviral and healing wounds in the form of slowly resorbable plates containing human leukocyte interferon for the treatment of local inflammatory diseases, including viral etiology.

Created drug needsto convenient for outdoor use and is applied by imposing on inflamed wet surface.

The offered plate is applied on the affected area once a day.

Slow resorption sponge provides the prolongation of action of medicinal substance interferon, immobilized on collagen.

Antiviral plate with interferon according to the invention is prepared as follows.

Example 1. In the mixer with the mixer load 250 g of 2% aqueous acetic acid solution of collagen and 200 g of water for injection, and then while stirring add 50 ml of concentrated interferon containing 33 thousand ME antiviral activity in 1 ml of the Mixture is bottled under aseptic conditions on kootam with the thickness of a layer of 6 mm and transferred to the camera freeze installation, such as TG-50. Freezing leads to a temperature of minus 25oC, then subjected to vacuum to vacuum of 0.1 mm RT. Art. and sublimation of the solvent at the temperature of the capacitor minus 50oC. the temperature in the final stage comes to the 28oC. the Total duration of the drying 20-24 hours. The sheets wound healing plate is cut into plate size 20 x 10 and 30 x 40 mm and is sealed in plastic bags and sterilized by irradiation with a dose of 15-25 kGy.

Example 2. In the mixer with the mixer load 250 g 2%-dobavlaut 100 ml of concentrated solution of interferon, containing 33 thousand ME in 1 ml. Next, the process of obtaining funds were carried out as in example 1.

Obtained according to the invention, the plate is examined for the presence of antiviral activity on tablets of human culture cell line spew against 100 JRS vesicular stomatitis virus. Transplantable cell culture spew displayed in 1960 K. S. Kulikova in laboratory cell cultures of the Moscow research Institute for viral preparations from kidney fetal pig. The antiviral activity was carried out before irradiation and after irradiation.

The results are presented in the table.

As can be seen from the table, the plate prepared according to examples 1 and 2, has a pronounced antiviral activity (14 and 20 thousand IU/plate). When sublimation activity of interferon is reduced to 3.5 times, similar to human leukocyte interferon dry, available in capsules.

Sterilization dose 15-20 GSR does not reduce the activity of the dried plate.

The claimed pharmaceutical composition can be applied locally on any site in need of treatment. The method of local treatment of the affected area provides the possibility of applying the proposed funds directly to the affected plot is about immunomodulation. -Immunology. - 1987, 4, S. 30-34.
