The method of obtaining the extract of the plant materials on the basis of walnuts

 

(57) Abstract:

The invention relates to pharmaceutical industry and veterinary medicine. Plant material is crushed fruits walnuts, oculoplasty and leaves with flower pollen and propolis extracted with two-stage jet fuel in the ratio of 1:1. The process is carried out at a temperature of 20-30oWith a length of 15-25 days in the first stage and 25-35 days in the second stage. The invention allows to increase the content in the extract biologically valuable components.

The invention relates to pharmaceutical industry and veterinary medicine, particularly to a method of obtaining the extract based on the nuts.

It is known that the fruits, bark, oculoplasty, walnut leaves contain biologically active substances, alkaloids, proteins, essential oils, vitamins, colorants (pigments plant) [1, 2]. Decoctions, tinctures, extracts of ripe and milky-wax ripeness walnuts, leaves and bark are used in folk and scientific medicine for wound healing, treatment of colds, skin, cancer [2, 3].

Additional processing of kerosene adsorbents increase the complexity of technology and the increase in the cost of preparation of the extract [5].

By the third method [3] crushed to 1 mm walnuts milky-wax ripeness is extracted in 2 stages. The first stage is mixed with oil distillation fraction "Liner" with low content of aromatic hydrocarbons in a ratio of 1:3, intensively stirred for 7 days, then aged 5 days at a temperature of 35-40oC. Extract t 10 days under the same conditions. Both the extract is separated, combined and passed through the dried residue (bagasse). The disadvantages of the method are: the complexity of the technology, including difficulties with the separation of the extract and residue in finely divided form, the possibility of adsorption of the biologically active components of the extract of the dried residue, high energy costs associated with the subdivision.

On the fourth way [6] the extract obtained from walnuts milky-wax ripeness in aviation kerosene (GOST 10227-86) at a ratio of 1:1 in two stages, with the crushing of the fruit of the walnut to the size of 5-7 mm, using fruits, okoloplodnika and leaves at a ratio of 1:0,7:0,3 with periodic stirring at a temperature of 30-35oWith duration 35-45 days at each stage. The disadvantages of the method is insufficient content of biologically active components.

The task of the invention is to increase the biological value of the extract.

This task is achieved by the fact that, as the plant materials on the basis of already used walnut raw material is crushed to a size of 5-7 mm fruit nut in oculoplastic and leaves at a ratio of 1:0.3 to use additional pollen (GOST 28887-90) and propolis (TC 10-01.2-92). AND THE P> Pollen contains valuable biologically active components is not less than 21% crude protein, not less than 2.5% of flavonoid compounds. Propolis also contains biologically valuable components is not less than 25% of the amount of phenolic compounds. Thus, the extract is transferred additional number of biologically valuable components of chlorophyll and other extractable chromotropic components, flavonoids, protein, phenolic compounds possessing anti-inflammatory and antibacterial action. The extraction is carried out in 2 stages at mass ratio of 1:1 at each stage at a temperature of 20-30oWith periodic mixing (once a day) to ensure the best conditions of extraction. In the first stage extraction should be performed within 15-25 days, on the second - 25-35 days. Extracts separated by decantation and unite.

The content of biologically valuable (active) components characterized by the following General quality indicators:

appearance - thin liquid from yellow-green to light-brown color;

density (at 20oC) - 0,78-0,86 g/cm3;

acid number (K. H.) - 0.2 to 0.5 mg KOH/g;

mass fraction of nitrogen by Kjeldahl method of 0.4 - 0.6% (in terms of pest (pigments) - 3-6,5 mg%.

According to a previously described method [5, 6] extract had the following quality indicators:

appearance - thin, liquid, yellow-green;

density (at 20oC) - 0,75-0,85 g/cm3;

acid number (K. H.) - 0.37 to 0.52 in mg KOH/g;

mass fraction of protein - 2,9%;

mass fraction of carotene - 0,37-0,52 mg%;

mass fraction of chlorophyll - 0.12 to 0.30 mg%.

In the proposed method increased the bioavailability of the drug: the drug contains phenolic compounds (positive phenols with ferric chloride), increased protein content, chlorophyll and other chromotropic substances.

Example. In a glass bottle with a capacity of 20 l load 10 kg crushed to a size of 5-7 mm of the nut in oculoplastic milky-wax ripeness with leaves in the ratio of 1:0.3 to (walnut raw materials), then 0,15-0,20 kg pollen, 0,12-0,18 kg of propolis and 10,33 kg of aviation fuel ratio (mass raw material : kerosene 1 : 1), stirred and maintained at a temperature of 20-30oC for 15-25 days on the 1st stage with periodic stirring, 1 time/day, the extract is separated by decantation, again download the same amount of kerosene and under the same conditions insist 25-to-green color with the following quality indicators:

density at 20oFrom 0.80 g/cm3, an acid number of 0.2 mg KOH/g;

mass fraction of carotene 0.3 mg%;

chlorophyll and other chromotropic compounds of 6.5 mg%, protein 3.1 per cent;

phenolic compounds (qualitative reaction for chlorine gland).

Thus, under the proposed method the extract is higher biological value.
