The invention relates to biotechnology, Microbiology, remediation of contaminated land. Received a new strain of Actinomyces sp. 1-96A analytical method selection of natural isolates on the principle of the degradation of petroleum products. Strain decompose the tar and oil in the culture fluid, containing 5% within 10 days. The strain can be used as remediate when cleaning surfaces and areas contaminated by petroleum products. table 2.

The invention relates to biotechnology and can be used when biostable soil and water contaminated with petroleum products.

Known strains used for purification of water and soil from oil and oil products, for example Rhodococcous erythropolis [1] oxidizes the oil at low concentrations: within 40 days, the oil content is reduced from 3 to 1%.

Known strain of Acinetobacter oleovarum [2], oxidizing aromatic hydrocarbons.

The strain of Actinomyces sp. 1-96A obtained by the method of stepwise analytical selection. Isolate isolated from soil samples of the Yaroslavl refinery, Tutaev). The strain is stored in kollektsii JSC "Bioflora", N B-05. The strain of Actinomyces sp. 1-the flora, has a collection of N B-05.

Cultural and morphological characteristics.

On solid (agar) medium of čapek, having the composition: sucrose 30 g; NaNO32 g; K2HPO41 g; MgSO47H2O - 0.5 g; KCl, 0.5 g; FeSO40.01 g; agar 20 g; distilled water 1000 ml, pH of 7.2, t = 291oC, forms a whitish aerial mycelium, on 3-4-e day highlights grayish-brown substance. Shporonosni long, slightly twisted in a spiral; spores oval and spherical. 8-10-e day on the surface around the colonies observed crystal-like formation, with side lighting gives Shine. Similar characteristics are observed when grown in the medium of čapek containing organic nitrogen.

Cultural and morphological characteristics do not change when grown on agar medium of čapek in the range from 12 to 29oC.

Physiological and biochemical characteristics.

Proteolytic activity is weak, gelatine slowly thins, milk rolls poorly, well hydrolyzes starch. Absorbs organic and mineral nitrogen, grows on fats, paraffin, naphthenic and asphaltene, the slow growth of the tissue.

In liquid medium N 65 DSM, having the composition: glucose, 4 g; progreesive with the addition of Wednesday 1-0.1% tar or oil. On the 3rd day of fermentation at 291oC on a rocking chair n = 180 rpm-1there are traces of the introduced hydrocarbon, the culture fluid is viscous, with the formation of bright flakes when defending (t = 4, 10oC).

The strain is maintained on agar nutrient medium N 65 with the addition of tar at 0.1%.

The strain can be used for remediation of areas and surfaces contaminated with heavy fractions of petroleum.

Example 1. The strain of Actinomyces sp. 1-96A were grown in liquid nutrient medium of čapek with the use of mineral nitrogen. Before sterilization in a flask with a nutrient medium made tar in an amount of 5%.

As a prototype grew Acinetobacter oleovarum BWA-712 liquid nutrient medium having the composition: (NH4)2SO4- 3.5 g; KH2PO4- 6.5 g; MgSO4- 0.7 g; FeSO47H2O - 12.5 mg; MnSO45H2O - 12.5 mg; ZnSO47H2O - 12.5 mg; CuSO4H2O - 3 mg; distilled water - 1000 ml Before sterilization was added into the medium tar in an amount of 5%.

The results of the experiment are presented in table. 1.

Example 2. The strain of Actinomyces sp. 1-96A were grown in liquid nutrient medium N 65 DSM, having the composition: glucose, 4 g; yeast extract, 4 g; ut in an amount of 5%.

As the prototype used a strain of Acinetobacter oleovarum BWA-712 liquid nutrient medium (see example 1). Before sterilization in a flask with a nutrient medium made oil in an amount of 5%.

The cultures were grown at 291oC on a rocking chair (n = 180 rpm).

The results of the experiment are presented in table. 2.

Thus, it was found that a strain of Actinomyces sp. 1-96A is promising for use in remediation areas and surfaces contaminated with oil.
